Jun 8, 2023 Researchers discovered a novel method to activate G-protein coupled receptors from inside the cells, and it helps develop drugs without side effects. June 08, 2023 Have you ever wondered how drugs reach their targets and achieve their function within our bodies? If a drug molecule or a ligand is a message, an inbox is typically a receptor in the cell membrane. One such receptor involved in relaying molecular signals is a G protein-coupled receptor GPCR. About one-third of existing drugs work by controlling the activation of this protein. Japanese researchers now reveal a new way of activating GPCR by triggering shape changes in the intracellular region of the receptor. This new process can help researchers design drugs with fewer or no side effects. If the cell membrane is like an Oreo cookie sandwich, GPCR is like a snake with seven segments traversing in and out of the cookie sandwich surface. The extracellular loops are the inbox for messages. When a message molecule binds to the extracellular side of the receptor, it triggers a shape change activating G proteins and β-arrestin protein attached to the intracellular side of the receptor. Like a molecular relay, the information passes downstream and affects various bodily processes. That is how we see, smell, and taste, which are sensations of light, smell, and taste messages. A new way to activate G protein-coupled receptors from inside the cell. Adverse side effects ensue if drugs acting on GPCRs activate multiple signaling pathways rather than a specific target pathway. That is why drug development focuses on activating specific molecular signal pathways within cells. Activating the GPCR from inside the cell rather than outside the cell could be one way to achieve specificity. But until now, there was no evidence of direct activation of only the intracellular side of GPCRs without the initiations from the extracellular side. A team of researchers headed by Osamu Nureki, a professor at the University of Tokyo, and his lab, discovered a new receptor activation mode of a bone metabolism related GPCR called human parathyroid hormone type 1 receptor PTH1R without signal transduction from the extracellular side. “Understanding the molecular mechanism will enable us to design optimal drugs,” says Kazuhiro Kobayashi, a doctoral student and an author of the study. Such a drug offers “a promising treatment for osteoporosis.” Kobayashi has been conducting research on bone formation in animal models since he was an undergrad. “Treatments for osteoporosis that target PTH1R require strict dosage, have administration route restrictions, and there aren't yet any better alternatives,” he says. That motivated their team to look for better drug design strategies targeting the parathyroid hormone receptor. To understand function through structure, they used cryo-electron microscopy and revealed the 3D structure of the PTH1R and G protein bound to a message molecule. The team synthesized a non-peptide message molecule called PCO371 which binds to the intracellular region of the receptor and interacts directly with G protein subunits. In other words, PCO371 activates the receptor after entering the cell. The PCO371-bound PTH1R structure can directly and stably modulate the intracellular side of PTH1R. And because PCO371 activates only G protein and not β-arrestin it does not cause side effects. This specificity of its binding and receptor activation mode makes it a suitable candidate for potential small-molecule-based drugs for class B1 GPCRs, like PTH1R, which currently lack oral administrative drug ligands. Such drugs would have reduced adverse effects and burdens on patients as they act on specific molecular pathways. The findings from this study will help “develop new drugs for disorders such as obesity, pain, osteoporosis, and neurological disorders.” The study appears in the journal Nature. Selain sebagai motivasi, tentu saja kita juga bisa mendapatkan Letter of Acceptance LoA dari Proffesor yang kalian minati. Langkah inilah yang mengantarkan saya ke Todai. Sejak Juni 2017, saya telah mencari dan mengeksplor siapa saja calon pembimbing yang sesuai dengan latar belakang, minat dan bidang saya. Akhirnya saya menghubungi beliau via e-mail. Calon pertama dan kedua tidak bisa karena habis masa kerjanya di University of Tokyo. Namun, beliau menyarankan saya dosen lain yang sekarang menjadi pembimbing saya. Beruntungnya, calon pembimbing saya ternyata sednag ada kegiatan di Jakarta, hingga saya dapat bertemu langsung dengannya di Bogor dan saya mendapat LoA sekitar satu bulan kemudian. Singkatnya, kita perlu jelas menentukan minat dan latar belakang dengan jurusan yang akan diambil. Selanjutnya, cari informasi sebanyak-banyaknya siapakah profesor yang paling sesuai dengan minat dan latar belakang kita. Ingat, hubungi satu profesor terlebih dahulu. Jika belum membalas, silahkan di-email kembali dan apabila menolak, tanyakan apakah beliau punya saran profesor yang sesuai dengan kita. Kuncinya di sini adalah tetap berusaha dan berpikir positif. Jangan lupa, saat menghubungi calon profesor, kalian perlu menyisipkan biodata CV atau Portofolio dan Research Plan kita. Selanjutnya, saya mencari informasi pendaftaran kuliah di Todai. Di Todai pendaftaran dilakukan melalui sistem T-cens Ikuti petunjuk dan baca dengan baik perintahnya, berikutnya ikuti langkah-langkah di dalamnya hingga seluruh proses selesai. Proses ini akan memakan waktu cukup lama kurang lebih satu tahun, namun pengisian form di dalam sistem akan dilakukan secara bertahap. Informasi baiknya adalah di dalam sistem ini terdapat informasi apa saja beasiswa yang dapat diikuti diantaranya ADB-JSP, Japan-Taiwan Exchange Association scholarship, JICA-FRIENDSHIP Scholarship Program IITH, Asian Future Leaders Scholarship Program, Todai-IIT Graduate Student Scholarship Program, dan Indonesia Endowment Fund for Education LPDP. Dengan kata lain, beasiswa tersebut sudah masuk dalam sistem T-cens. Program Beasiswa ADB-JSP Foto Bersama ADB-JSP Scholar of Urban Engineering Seperti yang saya sebutkan sebelumnya, ADB-JSP dan JICA telah masuk dalam sistem T-cens. Oleh sebab itu, pada langkah awal dalam sistem T-cens, Saya hanya perlu mencamtumkan beasiswa yang akan diambil, yaitu ADB-JSP dan JICA. Kita bisa memasukkan maksimum 3 pilihan di dalam sistem. Sebagai informasi, sebelumnya saya telah mendaftar beasiswa INPEX. Sayangnya, saya tidak mendapatkan kesempatan mendapat beasiswa ini. INPEX mengumumkan pada Rabu, 24 Januari 2018. Dan tepat sehari setelahnya, Kamis 25 Januari 2018, calon professor saya menghubungi bahwa saya menjadi short list kandidat penerima beasiswa JICA. Bukan hanya itu, sehari setelah itu, 26 Januari 2018, saya dihubungi oleh bagian administrasi departemen Urban Engineering atau dikenal FSO UBE bahwa saya menjadi short list kandidat penerima beasiswa ADB-JSP. Nikmat Allah luar biasa bukan? Ringkas cerita, proses seleksi beasiswa ADB-JSP dilakukan terlebih dahulu melalui sistem T-cens dan sampai akhirnya pada 27 April 2018, informasi penerimaan beasiswa ADB-JSP diumumkan dan saya menjadi salah satu penerimanya. Sangat lega rasanya mengetahui hal tersebut. ADB menawarkan beasiswa jenjang magister untuk negara berkembang yang menjadi peminjam dari ADB. Program beasiswa yang disebut ADB-JSP ini melingkupi bidang ekonomi, manajemen, sains dan teknologi, dan kajian pembangunan. Indonesia sebagai salah satu negara anggota dari ADB masuk dalam daftar negara yang bisa mendaftar untuk program ini. Dari program ini, telah lahir banyak alumni yang mengabdi di tanah air. Program ini membuka kesempatan hingga 150 orang untuk berkuliah jenjang master di universitas-universitas di berbagai negara. Kalian bisa melihat informasinya di Banyak sekali manfaat yang diberikan oleh beasiswa ini. Beasiswa ini memberikan biaya kuliah penuh, tunjangan bulanan untuk pengeluaran termasuk perumahan, buku dan materi pengajaran, asuransi kesehatan dan perjalanan. Bukan hanya itu, bagi para mahasiswa yang akan melakukan penelitian tersedia biaya khusus di dalamnya. Sebagai penutup, saya ingin menyampaikan pesan kepada seluruh pembaca. Pertama, rencana perlu dibuat sebelum melakukan sesuatu layaknya sekolah di luar negeri dan tidak semua rencana berjalan sesuai kehendak kita. Oleh sebab itu, kita perlu improvisasi dan membuat rencana dengan matang, serta menyiasati berbagai kemungkinan termasuk menyusun rencana B, C dan rencana tidak terduga lain. Seiring dengan berjalannya waktu terkadang motivasi turun dan naik, jadi usahakan selalu mengingat rencana dan impian dibalik itu semua, kenapa kita memilih jalan ini dan itu, serta membaca atau berinteraksi dengan orang-orang yang kita anggap memberikan dampak positif terhadap diri kita. Seluruh usaha kita perlu diiringi pula dengan doa dan dukungan keluarga. Selain menambah motivasi, hal ini juga menambah kepercayaan diri kita. Dan yang terakhir adalah penerapan ilmu yang akan kita dapatkan pada masyarakat dan kehidupan secara umum. Ini adalah tujuan yang paling penting.
